SomesclerodermapatientsproducehightiterantibodydirectedagainsthumantopoisomeraseI(Sheroetal.,1986). Theimmunoglobulinfractionwaspurifiedfromthesepatientsandshowntoreactspecificallywiththe100kDaformofTopoIusingWesternblotting. Theimmunecomplexesproducedwiththisreagentwillbind[125I]-proteinA. WhenintacttopoisomeraseIisused,asinglepolypeptideof100kDawasdetected;however,sub-bandsproducedbyproteolysiswillalsobindthisreagent,asexpected. TheantibodyisparticularlyusefulasaWesternblottingprobeandcanbeusedathighdilutions(>1:5,000). Crossreactivityofthisantibodywithotherspecieshasbeendemonstrated(withcalfthymus). Importantnoteaboutproductuse:HumantopoisomeraseIundergoesspontaneousdegradationespeciallywithrepeatfreezing/thawing.Theactivityoftheenzymeisnotaffectedsignificantly;however,sub-bandsmaybedetectableinWesternblottingexperiments.Thisantibodymaybeusedinimmunoprecipitationexperiments.
